Im Rahmen des BMBF-geförderten Drittmittelprojekts REA haben wir marine Enzyme (Peroxidasen) aus Makroalgen identifiziert, um neue Tools für die Bioindustrie zu generieren. Viele Peroxidasen und andere pflanzliche Proteine weisen ein hohes Potenzial für eine Kommerzialisierung in der Bioindustrie auf, jedoch nur in hoher Reinheit und Ausbeute, was sich durch eine Produktion in spezialisierten Organismen realisieren lässt (rekombinante Expression). Bakterien als Standardproduzenten von Proteinen haben aufgrund diverser post-translationalen Modifikationen von Pflanzenproteine häufig Schwierigkeiten, diese funktional aktiv zu bilden. In diesem Projekt Alga-Prot nutzen wir mit der Mikroalge Chlorella vulgaris einen bisher wenig verwendeten Expressionsorganismus. Da das Muster post-translationaler Modifikationen in Mikroalgen und höheren Pflanzen sehr ähnlich ist, ist der Ansatz vielversprechend, lösliche, aktive Pflanzenproteine rekombinante in Chlorella vulgaris zu produzieren. Im Rahmen dieses Pilotprojekts werden wir mit GFP zunächst ein „einfaches“ Testprotein produzieren lassen und eine Prozessentwicklung in Richtung der rekombinanten Produktion der MeerrettichPeroxidase HRP und alternativer Peroxidasen aus dem REA-Projekt starten.
